Monoclonal antibodies were generated in C57 wild-type mice (immunized with human αIIb transgenic mice platelets) or human αIIb transgenic mice (immunized with C57 wild-type platelets). Subtype and binding to platelets were analyzed by flow cytometry using washed human/mouse platelets pre-incubated with 5 μg/mL of monoclonal antibodies and then incubation with FITC-conjugated anti-mouse IgM, IgG, IgG1, IgG2a, IgG2b, and IgG3 secondary antibodies.
